A Protocol for the Optimization of Lateral Flow Immunoassay Strip Development

Background: Lateral flow immunoassay (LFIA) is a well-known method because of its extensive application in many fields, such as medical, food safety, and environmental analysis. LFIA also under development for pathogen or protein detection, where rapid detection essential. However, there are few studies on the optimal process production. Methods: This study aimed to evaluate roles different materials that make up strip, consisting sample pad, conjugate nitrocellulose membrane, absorbent pad. Different membranes pads were regarding membrane permeability, generation test line, capillary time, result visibility. Results: The results indicated optimized strip composed four pads, including CF3 FF170HP immobilized with 1 mg antibody, Standard 17 CF6 was visually detectable within 15 minutes. Conclusion: optimization protocol could be reference new strips.
